SPIRITT: a study of second-line treatment of metastatic colorectal cancer with FOLFIRI plus panitumumab or bevacizumab

Regimens incorporating irinotecan, oxaliplatin, capecitabine, and bevacizumab have significantly improved survival in patients with metastatic colorectal cancer (mCRC). Most patients eventually progress, however, requiring second-line therapy. Inhibitors of the epidermal growth factor receptor (EGFR) have demonstrated efficacy in mCRC for certain patients. Panitumumab, a fully human monoclonal antibody, has demonstrated activity as a single agent in the treatment of mCRC, and clinical study data indicate some activity in combination with chemotherapy. SPIRITT (Second-line Panitumumab Irinotecan Treatment Trial) is an open-label, randomized, phase II study evaluating 5-fluorouracil, leucovorin, and irinotecan, administered every 2 weeks, in combination with either panitumumab or bevacizumab as second-line therapy for patients with mCRC after the failure of first-line treatment with oxaliplatin-based chemotherapy plus bevacizumab.
