AMG 479 in combination with exemestane or fulvestrant in postmenopausal women with locally advanced or metastatic breast cancer

Although conventional hormonal therapy is effective for patients with estrogen receptor-positive metastatic breast cancer, virtually all patients with metastatic disease will eventually become resistant to conventional hormonal agents and develop progressive disease. Available second- and third-line therapies are less than ideal, producing only modest therapeutic gains. This phase II study of AMG 479 in combination with exemestane or fulvestrant is currently enrolling postmenopausal women with hormone receptor-positive, locally advanced or metastatic breast cancer whose disease has progressed on conventional hormonal therapy. AMG 479, a targeted agent in early-stage clinical development at Amgen with an overall favorable side-effect profile, inhibits the type 1 insulin-like growth factor-receptor (IGF-1R) pathway involved in tumor-cell proliferation and growth. The study will determine whether AMG 479 can sensitize tumor cells to respond to second-line hormonal therapy.
